Post subject: Immuohistochemistry of adipose tissue Reply with quote
Hi all,
I would like to do macrophage staining in sheep adipose tissue. I do not know where to start from? Any suggestions please !!!!!!!!!!

I recall this:
process to Pwax. Cut sections. Float out. Mount onto Superfrost Plus slides ( or your own APES-treated/subbed slides). Stick in 60C oven for 1hr minimum ( I usually leave overnight)
Dewax, rehdrate,H2O2 block ( if doing Px IHC), perform whichever HIER works for you.
Wash in tapwater.
Block in buffer containing BSA.
Incubate in Iba1 ( from Wako)
Detect with any system you wish.
Will not fail.
A general stABCpx-DAB method is in Protocols
