Hello,
I am looking for some general advice. I'm trying to section EDL (mouse) flash frozen tissue (stored at -80) at 16 and 30microns with limited success. The samples are shearing horribly. I've tried adjusting the temperature with, sharpened blades etc..
Unfortunately my advisor isn't great when it comes to muscle sectioning so I'm left combing the internet at all hours looking for some tips.
Anything would be greatly appreciated!

I missed this.
I am sure that you have sorted your problem by now?
Very difficult to troubleshoot such a problem over the web.
Freeze an easy tissue, like liver...cut it the same way.
Cut at 10 microns.
If that doesn't cut, you start looking at knife sharpness, temp of chamber.
Please let us know if you fixed the problem...
