Post subject: IHC detection system

| Hi I am using DAKO Envision as detection system. It is good for me but expensive. Can anybody suggest me an alternate detection system? Anybody has idea about Vector Lab? |

no suggestions.
I am surprised.
I think that most IHC kits are quite expensive ( kits= those containing RTU reagents + chromogen).
Vectorlabs is cheaper than Dako.
I use secondary biotinylated and stABCpx.
Much cheaper than polymer kits.
However, not as sensitive.
With micropolymer kits, one can dilute the primary Ab at least x2 more than with stABCpx.
So saving money on expensive primary Abs.
And time....
However, imho, it is the sensitivity of the DAB ( enhanced) that gives increased sensitivity, not the micropolymer reagent. |

| What is your opinion on Lab Vision? In our country only DAKO and Lab Vision are available. They have a detection system Ultravision Labeled Polymer Value Detection System Cat no TL-125-HLS. I also seek your advice regarding the c-erb MS-325-B and EGFR MS-378-P of Lab Vision. Our vender can not answer our question. Thanks |

I have not used labvision kits but, surely many readers have?
Can anyone comment?
I am sure that they are good kits as Labvision are an excellent Supplier.
I have not used the 2 Abs you ask about....you do not state what conditions you wish to use them under....
I hope Ole, Hogne or anyone else can comment.

The labvision kits i have used have been good. I agree with Carl "labvision as an excellent supplier".
I have not tried MS-325-B or MS-378-P.

MS-325 have not got an optimal score yet - but numbers are low for this one so its difficult to predict its quality.
Even tho an antibody get an optimal score for Her-2 its easy to overstain and understain the target protein. If you got good controlle tissues or a reliable ISH technique, its usually few problems getting and maintaining optimal staining if is a good antibody.

Seems to be the same results at Nequas for the few using clone e2-4001 - it fail to stain optimal. If you have good in-house controls (0+,2+ and 3+) for Her-2 and a HER-2 in situ hybridisation method for comparing results, its should be no problem to try/test the antibody (Ms-352) - it might be good?
Altho i would choose A0485 DAKO or SP3 Labvision. I use both of them and they perform great. Her-2 overexpression occur in 17-20% of the breast carcinomas and if your results give higher or lower results theres something wrong in the interpretation of results or the protocol. As mentioned its easy to overstain and understain this protein. If you can make TMAs - 50 breast carcinomas would do the evaluationprocess easier.
As for EGFR - i dont know. I have tried a few EGFRs over the years - we rarly use the antibody - in research occationally. I use EGFR, clone 31G7 (Zymed).
