Post subject: What is the best "negative" control for radioactiv
I've normally been using sense-strand probes as a negative control, with the aim of producing a blank section post-hybridisation. However with some recent work which showed a signal on my sense slides, I looked into another method- using a cold excess combination of labelled and unlabelled probe.
Here, I am mixing 100-150X cold probe with the same counts I would normally be applying to a section. My original hope was to again, produce a clean slide post-hybridisation, however at best, what I am seeing is only a fainter (in some cases, much fainter) signal than with anti-sense probes alone.
